中文摘要: 何首乌主要活性成份二苯乙烯苷(TSG)已被证实可通过调节多种靶分子起到抗动脉粥样硬化(AS)的作用。近期通过2-DE和MALDI-TOF-TOF等方法在AS大鼠主动脉组织中发现TSG对波形蛋白的表达及裂解具有调控作用。本研究在此基础上,从不同细胞水平(EC损伤凋亡模型﹑VSMC增殖迁移模型﹑巨噬源性泡沫细胞模型),通过Western Blotting﹑Northern Blotting﹑RT-PCR﹑免疫荧光﹑siRNA等方法,证实假设:TSG通过对波形蛋白的调控(包括波形蛋白的表达、裂解及与其它黏附分子的相互作用),起到抑制细胞黏附﹑迁移﹑凋亡及泡沫细胞形成的作用,以阐明TSG抗AS的作用机制,同时为波形蛋白作为新的TSG作用靶分子提供依据。

英文摘要: 2,3,5,4′-Tetrahydroxystilbene-2-O-β-D-glucoside (TSG) is an active component extracted from Polygonum multiflorum, which has been shown to inhibit the atherosclerotic process through many kinds of targets. We have recently discovered that TSG regulated the expression and cleavage of vimentin in atherosclerotic rats by methods of two-dimensional electrophoresis and MALDI-TOF-TOF. In this study, Western Blotting, Northern Blotting, reverse transcription PCR, immunofluorescence and siRNA technology will be employed in different cells (including endothelial injury model, proliferation and migration model of VSMC, macrophage-derived foam cell model), to confirm the following hypothesis: TSG regulates the vimentin cytoskeleton (including vimentin expression, cleavage and interaction with other adhesion molecules) to inhibit cell adhesion, migration, apoptosis and foam formation. The study will clarify the mechanism of TSG in treatment of AS and provide the basis for vimentin using as a novel TSG target.
