中文摘要: COPD是临床常见病、多发病,已经造成沉重的社会经济负担。目前,COPD的治疗仍以药物治疗为主,但尚无治疗该病的特效药。项目组前期的研究显示:芪蛭皱肺颗粒具有改善COPD模型大鼠的气道炎症,缓解气道阻塞,改善小气道重塑等作用,但尚未完全揭示其效应机制。研究证实mAChR的激活在COPD的发生发展中起着重要的作用,与COPD气道炎症反应、小气道重塑、气道细胞增殖、气道阻塞等病理过程密切相关。据此,我们认为调控肺组织mAChR的表达可能是芪蛭皱肺颗粒防治COPD的分子生物学机制之一。鉴此,本项目在前期研究的基础上,以COPD模型大鼠为载体,拟采用激光共聚焦免疫荧光双标技术及现代分子生物学技术从调控COPD肺组织mAChR表达的角度解析芪蛭皱肺颗粒防治COPD的可能机制,冀为芪蛭皱肺颗粒防治COPD提供理论依据。

英文摘要: Chronic obstructive pulmonary disease (COPD) is one of the common and multiplex diseases in clinical practice. It has caused the heavy socioeconomic burden in patients with COPD. At present, medications is regarded as the main method for COPD, but no specifics is used to cure it. Our forepassed studies showed that Qizhi Zhoufei Granules could improve the airway inflammation and remoulding, and alleviate airway obstruction in mouse with COPD, but they could not completely promulgate acting mechanism of Qizhi Zhoufei Granules for COPD. Some studies indicated that the activation of mAChR plays the vital role in initiation and development of COPD, and involves inflammation, remoulding, obstruction and cell proliferation of airway. In view of the above, we hypothesize that regulating expression of mAChR is one of the molecule biology pathogenesis of Qizhi Zhoufei Granules for COPD. Because of all this, this study would further investigate the putative mechanisms of Qizhi Zhoufei Granules for COPD from the viewpoint of regulating expression of mAChR in lungtissue via the techniques of Laser confocal microscopy-immunofluorescence e-tagging (LCM-IFE), and would be expected to provide the theory evidence for Qizhi Zhoufei Granules to prevent and cure COPD.
