中文摘要: 骨组织的生物模拟不仅要复制天然骨的化学成分,更重要的是再现骨的基本分级结构。近年来,仿生学家发现,天然非胶原蛋白(non-collagenous proteins, NCPs)在骨组织基本分级结构的形成中起着重要作用。本课题组在前期研究中发现两种多聚分子,可分别模拟NCPs对羟基磷灰石(hydroxyapaptite,HA)微晶大小和排列方向的调控。本项目拟在前期研究的基础上,采用复凝法组装三维的I型胶原微球,利用NCPs模拟物对HA微晶的双靶向调控作用,实现纳米HA在胶原纤维内的有序排列,复制出与天然骨组织具一致分级结构的Col/n-HA 微球,并探讨其在牙槽骨再生中的应用。本项目有望发展一种新的高仿生纳米骨再生材料,为生物模拟研究提供新理念,为临床牙槽骨缺损修复提供新材料。

英文摘要: Biomaterials for bone regeneration should replicate similar components and hierarchy to natural bone.Recently,biomimetist has found that non-collagenous proteins (NCPs) play important role on the formation of structual hierarchy in bone. In our previous studies, two NCPs analogs were applied to control the size of hydroxyapatite(HA) at nanoscale and the way of HA depositing within collagen fibrils.We successfully recapitulate different levels of structural hierarchy in mineralized collagen at two-dimension.In this project,three-dimentional collagen micorspheres will be assembled using complex coacervation method and two NCPs analogs will be applied to control the size and arrangement of HA.We plan to investigate the mechanism that hierarchically-arranged Col/n-HA microspheres assemble in three-dimention and the application of this scaffold in alveolar bone regeneration.
