中文摘要: 前期采用高通量纳米微柱阵列技术建立了少突胶质前体细胞(OPCs)分化和髓鞘形成筛选平台,能够定量和高效评估OPCs分化和髓鞘形成。通过筛选,鉴定了以氯马斯汀为代表的七个胆碱能受体拮抗剂(MRA)能够高效促进OPCs分化和形成髓鞘。我们推测OPCs表达的五个胆碱能受体亚型(M1R-M5R)中的一个或多个受体,是MRAs的靶点,同时调控OPCs分化和髓鞘形成。本课题拟采用基因沉默技术,敲基因小鼠等方法,结合离体OPCs培养和在体观察,明确MRAs作用的受体。进一步采用高通量纳米微柱阵平台,结合目前筛选结果,尤其是利用靶向胆碱能受体下游的信号通路分子的激动剂和拮抗剂,明确该胆碱能受体亚型的作用机制及信号通路。本项目的完成将首次阐明胆碱能受体对少突胶质细胞分化和髓鞘形成的调控作用和机制,有望为脱髓鞘疾病的治疗研究提供新的药物靶标。

英文摘要: Our previous study has established a high-throughput screening platform by using micropillar arrays for oligodendroglia differentiation and myelination. Upon screening 1500 compound library, we identified a cluster with seven muscarinic receptor antagonists (MRAs) that can promote the differentiation and myelination of oligodendroglia. We propose that one or multiple of the muscarinic receptor subtypes (M1R-M5R) are the targets of MSAs and also regulate the differentiation of oligodendroglia. The goal of the proposal is to identify the subtype(s) of muscarinic receptors that display as the target(s) of MRAs by utilizing siRNA, knockout mice and in vitro culture system. And investigate the downstream of muscarinic receptor by analyzing the screening data and the antagonists and agonists in the compound library. This project will illustrate the regulatory effects and downstream mechanisms of muscarinic receptors on the differentiation and myelination of oligodendroglia, and may provide new therapeutics for demyelinating diseases.
