中文摘要: 海洋中存在数量巨大种类繁多的微生物,推动着海洋各物质循环尤其是氮循环的进行。但是人们对于参与海洋氮循环的微生物种群及其所分泌酶的了解非常有限。中国黄海是半封闭陆架海,海流和潮流比较复杂,沉积物中氮含量略高,使黄海蕴藏着丰富的细菌物种资源,它们是研究大西洋西海岸氮循环流动的理想区域。本项目拟自黄海近海水域中采集年度周期的表层海水和沉积物样品,从中分离产蛋白酶细菌,研究该区域产蛋白酶细菌的多样性和相关菌群随位点季节的变动;研究这些菌株所产蛋白酶的多样性及其在不同位点和季节的分布规律;对疑似细菌新种、属菌株进行系统分类学鉴定,建立多个细菌新属、种等新的分类单元;鉴定和发现1-2个具有新的氨基酸序列或新的催化特性的新型蛋白酶资源。该研究将为进一步阐明中国黄海近海有机氮的降解和氮循环机制奠定基础,发现和丰富黄海菌株资源和蛋白酶资源,为黄海微生物资源的开发利用提供新的途径和思路。

英文摘要: There are tremendous microorganisms with high diversity in the ocean, which promote material recycling especially marine nitrogen cycle. However, the diversity of microorganisms and their extracellular proteases involved in the marine nitrogen cycle remain largely unknown. China's Yellow Sea is semi-enclosed shelf sea where current and tide is comparatively complicated. The Yellow Sea is an ideal area for studying the nitrogen cycle in the west coast of the Atlantic because of the huge nitrogen content in the sediment and abundant microbial resources. In this research, we will collect the surface seawater and sediments of the Yellow Sea at different stations in different seasons and isolate protease-producing bacteria from these samples, in order to study the diversity of the protease-producing bacteria in this area and their distribution pattern in different stations and seasons. The phylogenetic taxonomy of the strains which suspected to be novel species or genus will be investigated to discover novel taxon of novel species or genus. One or two new type proteases with novel amino acid sequences or novel catalytic properties will be discovered and identified.
