中文摘要: 研究证实miRNAs参与脓毒症的多种生物学过程,其中我们一项研究证实血清miR-122含量同脓毒症患者短期病死率相关。在本课题中我们前期研究在临床样本中首次初步证实miR-122在脓毒症凝血障碍中具有促凝作用,接下来我们将(1)生物信息学分析miR-122预测靶基因在促凝过程中的可能靶点;(2)在细胞内探讨miR-122导致促凝作用可能靶点及对凝血通路的影响;(3)在动物水平证实miR-122水平变化对凝血通路组分及脓毒症动物模型病死率变化的影响。本课题将拟从细胞和动物水平揭示miR-122参与凝血过程的可能分子机制,为脓毒症过程凝血紊乱的治疗提供新靶点。

英文摘要: Researches have proved that miRNAs were involved in many processes of sepsis patients, and one research of ours has proved that levels of serum miR-122 were correlated with the short-term mortality of sepsis patient. In this research , we firstly proved that miR-122 correlated to the coagulation disorders of sepsis patients. Then, we will be focused on (1) Bioinformatics analysis to predict possible target genes of miR-122 involved in the procoagulant process; (2)To explore the mechanism of miR-122 effecting the coagulation pathway in vitro;(3)To explore how the expression changes of miR -122 can effect coagulation pathway and mortality of sepsis in vivo. Our research is in order to reveal the mechanism of miR-122 invovled in coagulation process of sepsis patients in vitro and in vivo and will provide novel treatment target for coagulation disorders of sepsis patients.
