中文摘要: 丝状真菌是纤维素酶主要的工业生产菌株,然而丝状真菌中纤维素酶表达调控网络的分子机制非常复杂,远远没有阐释清楚。近期我们通过筛选模式纤维素降解真菌粗糙脉孢菌C2H2锌指转录因子突变体库,发现缺失一个新转录因子(命名为CEA-1)将显著降低纤维素酶的表达水平。本项目将重点针对转录因子CEA-1,通过分子遗传学、转录组学等手段,确定CEA-1下游调控基因及其作用位点,同时结合CEA-1与已知关键转录因子组合突变体和过表达突变体,解析CEA-1与已知调控元件的相互关系。CEA-1作为脉孢菌纤维素酶系表达调控网络中的新元件,其分子功能的解析,对阐明丝状真菌纤维素酶系表达调控的机理、提高工业菌种发酵水平都有着重要的意义。

英文摘要: Filamentous fungi are the major microbial producers of lignocellulolytic enzymes in industry. However, mechanism of gene regulation of cellulase expression in filamentous fungi is still far from being completely understood. We recently found a novel regulatory factor (named as CEA-1) affecting the production of cellulases by screening mutant library. So this project will focus on the regulation mechanism of this transcriptional factor. Compared the transcriptional profilings of WT and Δcea-1 grown on different induced conditions, the target genes of this transcriptional factor could be identified. And the binding motif could be aslo obtained. Meanwhile, the relationship between this factor and the other known factor genes invovled in lignocellulolytic enzymes expression could be elucidated. The characterization of this novel transcription factor could be beneficial for scientific understanding of the regulation mechanism of lignocellulolytic enzymes expression in filamentous fungi. It could also provide new cost-effective strategies for biomass degrading enzymes production.
