中文摘要: 昆虫与真菌是自然界最大的两类生物,长期协同进化形成了共生、寄生、捕食等复杂的互作关系。Attine蚂蚁、白蚁与菌圃真菌及小蠹虫与蛀道真菌等是真菌与昆虫互惠共生和协同进化的典型代表,具有复杂的互作机制。然而Euops属卷叶象甲与叶苞真菌作为另一类典型的共生协同进化关系,它们的这种共生关系维持机制尚未明确。本项申请拟通过Euops属卷叶象甲、叶苞及宿主植物叶片微生物群落分析,明确优势微生物类群;通过优势微生物类群及拮抗关系测定,明确形成共生关系的关键物种;通过共生菌原位杂交定位研究,明确其时空分布动态及协同进化关系;通过共生菌次生代谢产物及抗菌活性分析,揭示共生真菌与卷叶象甲协同进化的化学机制。研究结果不仅丰富真菌与昆虫协同进化理论体系,同时为重要的抗菌化合物发现提供新的资源。

英文摘要: Fungi and insects, as the two largest groups of organisms, have interacted for a long time and coevolved various relationships such as symbiosis, parasitism, predation etc. The coevolutionary and chemical mechanisms of ant-fungus mutualism and beetle-fungus mutualism have been well documented, while that of Euops weevils and their mycetangial fungi have not been well understood. The aims of current proposal are 1) to investigate the microbial community of the Euops weevils, leaf cradles and the host plant leaves; 2) to identify the symbiotic microbes in the mutualism relationship by detecting the antagonistic interactions between the dominant microbes and other microbes; 3) to observe and determine the distribution of symbiotic microbes on the Euops weevil, leaf cradle and host plant leaves by in situ hybridization localization analysis; 4) to identify the chemical mechanism of Euops weevils and symbiotic microbes by identifying the secondary metabolites and evaluate their anti-microbial activities. The results will not only provide comprehensive understanding co-evolution between fungi and insects, but also can lead to potential drug discovery.
