中文摘要: 多囊卵巢综合征(polycustic ovarian syndrome,PCOS)是育龄妇女中常见的生殖功能障碍性疾病,主要表现为卵巢的雄激素过多及无排卵。PCOS呈现高度的家族聚集性,提示其发病可能与遗传有关,但PCOS病人的体内环境变化也可能影响生殖细胞的表观遗传变化。本研究将通过注射脱氢表雄酮(dehydroepiandrosterone,DHEA), 建立小鼠PCOS模型,利用偏重亚硫酸氢盐测序法和COBRA等方法,研究高雄PCOS卵巢生理环境和代谢的变化是否影响卵母细胞的DNA甲基化印迹的正确建立以及印迹甲基化异常是否可通过生殖细胞传递给下一代,并利用人高雄PCOS病人卵子对小鼠的结果进行验证。从表观遗传学的角度解释PCOS导致卵子质量和生育力下降的原因和对胚胎发育和后代所带来的不良影响,揭示PCOS的发病原因在遗传上是否与DNA甲基化异常存在联系。

英文摘要: Polycustic ovarian syndrome (PCOS) is one of the major reproductive diseases that impair female reproduction and the infertile women often show excessive androgene and anovulation. The occurrence of PCOS is found to be transgenerationaly inherited, but whether internal environmental changes of PCOS patients causes germ cell epigenetic alterations that tansmit to the next generation is unknown.In the present study, we aim to establish PCOS mouse model by injecting denydroepiandrosterone (DHEA) and study the imprinting DNA methylation changes in oocytes and offspring germ cells using bisulfite sequencing and combined bisulfite restriction analysis (COBRA). The results will also be verified by using DNA imprinting analysis in human oocytes obtained from PCOS patients in IVF clinics.The data will explain whether DHEA-induced PCOS causes imprinting defects in oocytes and whehter such changes will be transmitted to the next generation. This study will reveal whether epigenetics plays a role in PCOS, in addition to genetic factors.
