中文摘要: “镜像骈体”是人工诱导棘尾虫表型变异,但其形成的分子机制尚不清楚。本项目拟采用表观遗传学的理论和分子生物学技术手段对镜像骈体发生的分子机制进行研究。分析棘尾虫无性生殖世代镜像骈体型与野生型转录组图谱差异,筛选选择性差异表达基因;分析选择性差异表达基因的时空动态表达图谱,鉴定基因功能,比对镜像骈体与野生型细胞皮质形态建成及细胞表型变化的异同,探讨候选基因的表达调控与细胞皮层形态建成的核遗传学基础。在上述研究基础上,研究参与镜像骈体细胞皮质形态建成的基因间或基因与蛋白质之间的相互作用关系,为探索调控镜像骈体发生的分子细胞信号通路、及为阐明真核生物细胞皮层发育的表观遗传调控机制提供理论依据。

英文摘要: “Mirror-imaged doublet” is phenotypic effect in Stylonychia mytilus protozoa form conjugation experiment. However, the molecular mechanism is not clear. The project intends to study the molecular mechanisms of mirror-imaged doublet using techniques of molecular biology based on epigenetics. Analyzing of difference of transcription profiling between mirror-imaged and wild type Stylonychia in asexual generation, and screening of differentially expressed genes; analyzing time-course gene expression profiles of differentially expressed genes, determining gene functions, comparing the similarities and differences on cortical morphogenesis and cell phenotype change between the mirror image doublets and wild type cells,and discussing genetic basis on the expression regulation of candidate gene and cell cortex forms built. Based on these studies, studying on the interaction between genes and proteins which participate in mirror image doublets cell cortex morphogenesis, in order to study cell signaling pathways for doublet occurring, clarify epigenetic regulation mechanism of the cell morphogenesis developmental in eukaryotic cell cortex.
