中文摘要: MRI增强检查是癌症早期诊断的最重要的手段之一。目前临床使用的MRI造影剂钆配合物在体内循环时间较短,不具有生物靶向作用,对肿瘤组织的成像、转移灶的早期发现以及鉴别复发肿瘤组织与手术疤痕的效果均不理想。临床急需改进现有的造影剂以便帮助广大医生做出更明确的诊断。超顺磁性的Fe3O4纳米粒子毒性小,生物相容性好且能够生物降解,是目前较有前途的理想的MRI造影剂。肿瘤细胞表面的ανβ3整合素可以与特异性靶向的精氨酸-甘氨酸-天冬氨酸短肽(RGD)结合,本研究拟利用这一特点,通过化学作用将c(RGDFK)标记到Fe3O4@SiO2纳米粒子表面,以使造影剂能够同肿瘤细胞特异性结合,在肿瘤部位聚集,提高肿瘤诊断及鉴别的准确性。同时我们将在非小细胞肺癌nude小鼠模型中,验证Fe3O4@SiO2-RGD磁共振纳米靶向造影剂的显影效果,及其对肿瘤转移病灶的敏感性,为寻找理想造影剂在临床上的应用打下基础。

英文摘要: Enhanced MRI is one of the most important means of early diagnosis of cancer. MRI contrast agents currently in clinical use of gadolinium complexes in vivo circulation time is shorter, and does not have a biological targeting. It tumor tissue imaging, early discovery and identification of metastatic tumor recurrence and surgical scars results are not satisfactory. Clinical need to improve the existing contrast.agents in order to help the majority of doctors make more definitive diagnosis. Superparamagnetic Fe3O4 nanoparticles toxicity, biocompatibility and biodegradable, is more promising ideal MRI contrast agents. Ανβ3 integrin binding surface of tumor cells can be specifically targeted with RGD. This study intends to use this feature, the c (RGDFK) mark to Fe3O4 @ SiO2 nanoparticles through chemical action. It is possible to make the contrast agent with tumor cell-specific binding, accumulation at the tumor site, tumor diagnosis and improve the accuracy of identification. We will in non-small cell lung cancer nude mouse model validation developing new MRI contrast agent effect,and the sensitivity of tumor metastasis. To lay the groundwork to find the ideal contrast agents in clinical applications.
