中文摘要: 提高品质是谷子育种的重要方向,分子育种在品质性状遗传改良方面较传统育种存在明显的优势。本项目以基因组测序材料豫谷1号不同发育时期的籽粒为材料,利用转录组测序的方法从mRNA、microRNA和长非编码RNA(lncRNA)三个切入点分析谷子籽粒发育进程中品质相关基因的表达与调控情况。主要目的是深入了解谷子籽粒品质形成的分子机制,为谷子品质分子育种奠定基础。研究内容包括:1、构建籽粒发育进程基因表达谱,注释差异表达基因与可变剪接;2、分析籽粒发育进程microRNA表达及差异表达microRNA对靶基因的调控;3、分析籽粒发育进程差异表达的长非编码RNA分析并预测功能;4、筛选谷子籽粒发育进程中物质积累、转化相关候选基因。本项目从转录组层面对谷子籽粒品质形成过程的分子机制进行整体研究,实验结果对品质相关基因的克隆、遗传转化、分子设计育种提供数据支持。对谷子品质育种、遗传改良具有重要意义。

英文摘要: Molecular breeding has advantages over the traditional breeding in quality improvement which was important area in foxtail millet (Setaria italica L.) breeding. In this study, inbred Yugu1, accession of reference genome sequencing, was selected to analyze the gene expression and regulation relate to quality during seed development using transcriptome sequencing. The expression and regulation of mRNA, microRNA, and long non-coding RNA (lncRNA) was analyzed in this experiment. The purpose of this study is to get insight into the molecular basis of quality accumulation process, and establish bases for molecular breeding of quality improvement in foxtail millet. The detailed designs of the experiments are as followed: 1) to build the gene expression profiles during seed development, and annotate the different expressed genes and alternative splicing. 2) To study the expression profiles of mircoRNA during the seed development, and the regulation of the different expressed microRNA. 3) To analyze the changes in lncRNA expression, and predict the functions. 4) To mine the candidate genes related to material synthesis and transformation.
