中文摘要: 卵转铁蛋白(OTf)是禽蛋的主要生物活性物质,具有免疫增强作用。树突状细胞(DCs)是机体主要的抗原递呈细胞,能刺激初始型T细胞的增殖分化,是机体免疫的始动者,然而OTf对DCs功能的影响及其免疫调节机制还不清楚。本项目拟建立肠淋巴DCs体外细胞培养技术,比较OTf对健康小鼠和免疫低下小鼠肠淋巴DCs发育、成熟等影响;检测其相关的表面MHC-Ⅱ和共刺激因子CD80、CD86、CD40,趋化细胞因子受体CCR7和CXCR4的mRNA表达和p38MAPK、ERK、JNK等相关蛋白质含量的变化;及对细胞因子TNF-α、IL-10、 IL-12、 MCP-1、MIP-1α、MIP-1β、RANTES等含量的影响。从细胞和分子水平阐明OTf对小鼠肠DCs功能的影响及其免疫调节分子机制。本研究可为OTf作为免疫调节功能因子在保健食品中的应用提供理论依据,对禽蛋的深度开发利用具有指导意义。

英文摘要: Ovotransferrin(OTf) is the main active component in eggs, and has many functions such as enhancing immune function. Dendritic cells (DCs) is the key antigen presenting cell, it can stimulate the proliferation and the differentiation of the initial T cell, and is the initiator of body immune, but the regulation function and mechanism of OTf on DCs is still unknown. This study, the intestinal lympha DCs cultural technology will be built in vitro, and the effects of OTf on intestinal lympha DCs preliferation and maturation will be investigated in normal and immunocomprised mice, also the surface MHC II and costimulators as CD80, CD86, CD40, mRNA expressions of chemotactic cytokines receptors CCR7, CXCR4, protein expressions of p38MAPK, ERK, JNK will be detected, also the effect of OTf on the level of cytokines as TNF-α, IL-12,IL-10,MCP-1,MIP-α,MIP-β and RANTES will be detected. The effect and molecular mechanism of OTf on intestinal mucosa function will be investigated at cellular and molecular levels. The results of the study will provide a theoretical basis for OTf as an immune regulator in functional food, and have a profound affect and an important acadimic value for eggs further utilization and development.
