中文摘要: 当今社会正步入老龄化阶段,对于衰老干预措施的研究已经成为一项全球性的社会和经济问题。中医药对衰老和衰老干预措施的研究历史源远流长,但多数缺乏确实有力的实验证据与机制研究。 左归丸为补肾益精的要药,是临床常用于抗衰老的经典名方。申请者在前期的研究中发现,左归丸可促进肾虚大鼠骨髓间充质干细胞(MSC)的增殖,影响其分化能力的改变。本课题依据"肾精亏虚"的衰老理论及"干细胞具先天之精属性"理论,运用细胞生物学、形态学、影像学、分子生物学等技术方法,进行体外及整体动物实验,从MSC形态学、增殖分化能力、移植修复能力的改变与细胞衰老相关基因及Wnt/β-catenin信号通路相关性分析等方面探讨左归丸延缓MSC衰老的可能机制,从干细胞角度研究补肾益精法抗衰老的作用机理。 为深入揭示中医药延缓衰老的机制研究提供创新性的思路,为左归丸应用于临床抗衰老的保健与治疗提供有力的实验依据。

英文摘要: Today's society is entering the aging stage, and the study of aging interventions has become a global social and economic problem. Traditional Chinese medicine has a long history of research on aging and aging interventions, but most of these studies are lack of really powerful experimental evidence and mechanisms research. The Zuogui Pill is a classic prescription of invigorating kidney and nourishing essence, commonly used in anti-aging clinically. In the previous study, applicants found that Zuogui Pill can promote the proliferation of bone marrow mesenchymal stem cells (MSC) from weak kidney rats, affecting these cells differentiation capacity. According to "essence and blood have the same source" and "stem cells have the same properties as congenital essence", in vitro and in vivo experiments, we use cell biology, morphology, imaging, and molecular biology techniques, to discuss the possible mechanisms of delaying aging of MSC by Zuogui Pill, and anti-aging mechanism of invigorating kidney and nourishing essence from the perspective of stem cell research.
