中文摘要: 在检测药物干预对实验动物血清中性激素、抑制素、抗苗勒激素等经典指标调控的基础上,采用代谢组学与高内涵筛选等新的研究手段,围绕卵巢颗粒细胞,从其形态、功能、内源性代谢产物变化的角度,探讨广东省著名妇科专家张玉珍教授的经验方加减归肾丸抗卵巢衰老的作用机制,并初步进行物质基础研究:(1)采用D-半乳糖制造小鼠卵巢衰老模型。(2)检测药物干预对小鼠血清中性激素、抑制素、抗苗勒激素水平等常规指标的调控。(3)进行卵巢颗粒细胞的代谢组学与高内涵筛选研究,明确细胞内源性代谢产物的变化,明确细胞形态、结构、功能等的变化,从卵巢颗粒细胞的层面深入阐释加减归肾丸抗卵巢衰老的作用机制。(4)采用UPLC/MS进行原药、入血成分有效物质基础研究,初步明确加减归肾丸的有效物质基础。(5)构建作用机理与物质基础网络,揭示内在联系,阐释加减归肾丸组方的科学内涵。

英文摘要: This project is focused on the mechanism and substance base investigation of the TCM formulation Jiajian Guishen Pills on ovarian failure, developed by Prof. Yuzhen Zhang who is a well-known gynecologic expert in Guangdong Province. The traditonal classical indicators will be employed such as sex hormone in serum, inhibin and anti Mullerian hormone. Further research will be carried out based on metabolomics, high content screening (HCS) and subtance base. (1) D-galactose will be used to establish a mouse model of ovarian failure.(2)Sex hermone, inhibins and anti Mullerian hormone in serum will be detected to reflect the adjustment activity of drug.(3)Investigation of metabolomics and HCS of ovarian granule cells will be performed to indicate the variation of endogenous metabolites as well as the variation of characterization, structure and function of ovarain granule cells. Based on the research on granule cells, the mechanism of anti ovarian failure of Jiajian Guishen Pills will be recovered.(4)UPLC/MS will be used to analyze the raw materials, components moved in blood to basicly reveal the substance base of Jiajian Guishen Pills for anti ovarian failure.(5)The basic network of mechanism and substance base will be established to reveal the scientification of Jiaian Guishen Pills on anti ovarian failure.
