中文摘要: 由流感病毒引起的流感,每次爆发都会造成死亡,引起恐慌,给社会带来巨大经济损失。虽有文献报道流感病毒NS基因编码的NS1和NS2蛋白与病毒致病性和毒力相关,但对它们的功能研究还很少。在前期研究中,申请人利用反向遗传系统将流感病毒模式毒株PR8 NS基因中编码NS1蛋白的基因全部或部分与新型流感病毒H17N10的对应基因进行替换,结果表明NS基因编码的起始10个氨基酸会影响流感病毒的包装复制。本研究将进一步确认影响流感病毒复制的关键氨基酸位点,探讨其影响病毒复制的机理,深入了解流感病毒的致病机制,为抗流感药物的研发或抗体的制备提供新的靶标或依据。

英文摘要: Each outbreak of influenza will cause death, panic and tremendous economic losses. Although some literatures reported that influenza virus NS1 and NS2 protein encoded from NS gene contributed to viral pathogenicity and virulence, the functional study of them is not much. In previous study, we exchange the NS1 sequence of PR8 and H17N10, which is a new strain of influenza A virus, fully or partly by reverse genetics plasmid, the result showed that the starting 10 amino acid affects the virus rescue. In this subject, we will confirm the critical amino acid further and explore the mechanism of its effects on viral replication, which will help us understand the influenza virus better and provide new clues for the development of anti-flu drugs or antibodies.
