中文摘要: 高分辨软X射线成像是近年发展起来进行细胞成像的有力工具,它无需切片可以获得细胞的三维图像,观察到细胞器的三维结构(如可以观察到质膜、核膜、核孔、内质网、溶酶体和线立体的结构等),可以研究细胞受病毒感染后细胞器结构的变化,为疾病引起的细胞结构变化机理研究提供了强有力的研究手段。但成像系统的分辨率越高,系统的焦深就越小(对软X射线成像系统而言,波长比硬X射线的波长长,焦深更短),因此成像细胞的尺寸就越小,所以软X射线成像系统难以实现大尺寸(如真核细胞)细胞的清晰成像,为此,必须解决成像系统分辨率与焦深的矛盾。本项目拟采用小波变换和光学传递函数方法实现高分辨软X射线成像系统的焦深拓展,实现高分辨软X射线成像系统的大细胞成像。

英文摘要: The transmission soft X-ray microscopy which is developed in recent years is a powerful tool to yield the three-dimensional structure of whole cell in their native state based on X-ray tomography.It allowed us to visualize subcellular structure such as the double nuclear membrane, nuclear pores, nuclear membrane channels, mitochondrial cristae and lysosomal inclusions etc. It is possible to detect change of cell structure to compare the images obtained from virus infected cell and control cell. The depths of focus decreases with resolution of soft X-ray microscopy improved. The thickness of cell is limited when the cell is imaged in the high resolution soft X-ray microscopy system. It is of vital importance to extend the depths of focus to image thicker cells. This project intends to adopt the wavelet transform and optical transfer function to expand the depth of focus of the high-resolution soft X-ray imaging system,so as to achieve high-resolution large-cell imaging.
