中文摘要: 循环肿瘤细胞(CTCs)是肿瘤细胞经血道转移时的特殊存在形式和必经阶段,是决定病人生存率和治疗方案的关键指标之一。然而原发肿瘤如何进入血循环成为CTCs、CTCs如何在血流环境中生存、适应以及最终出血管形成继发瘤,这一系列过程的机理尚不明确,缺乏有效的研究工具是制约相关研究的主要原因之一。本课题拟利用微流控技术构建并完善肿瘤细胞转移血流动力学体外研究模型,实现实时动态监测和干预CTCs在近生理、病理血流动力学环境下的行为及其相伴的关键分子事件的发生、发展,进而揭示其机理。本课题的实施将为肿瘤细胞经血道转移的机理研究提供新型技术平台并有望大大促进其研究进展及早期防治。

英文摘要: Circulating tumor cell (CTC) is a special form of tumor cells and an inevitable stage of the hematogenous metastasis. CTC is a key factor deciding survival rate of patents and therapeutic regimen. However, how primary tumor cells enter circulation system and become CTCs, how CTCs survival and adapt to blood stream environment and eventually extravasate to form secondary tumor are still obscure. The lack of effective research tools is one of major reason for this situation. In this project, we will design and perfect an in vitro model for studying metastasis of tumor cells in hemodynamic environment using microfluidic techniques. Using this model, we plan to realize real time observation and manipulation of the behavior of CTCs under near-physiological/pathological, hemodynamic conditions. At the same time, we will investigate the mechanism of key events took place during the tumor metastasis. The implementation of this project will provide novel technical platform for the research of hematogenous metastasis of tumor cells and will greatly promote early prevention and treatment of cancer.
