中文摘要: EB病毒(EBV)感染与恶性淋巴瘤的发生关系密切。前期研究中我们利用miRNA芯片技术从EBV诱发淋巴瘤模型中筛选到3个差异表达的EB病毒miRNAs,其中ebv-miR-BART16对宿主细胞凋亡信号通路具有调控作用,可能通过这一机制促进或诱导淋巴细胞恶性转化。本课题拟采用报告基因技术证实ebv-miR-BART16对凋亡信号通路相关基因的靶向调节作用,运用细胞模型、荷瘤小鼠模型检测评价ebv-miR-BART16对淋巴瘤细胞系增殖和抗凋亡恶性表型的影响,通过检测细胞凋亡及凋亡信号通路各级联分子的活性,获得ebv-miR-BART16参与调节细胞增殖与凋亡的直接证据,揭示ebv-miR-BART16的生物学功能及其在淋巴瘤发生中的作用机制。

英文摘要: Epstein-Barr virus (EBV) is closely associated with human malignant lymphoma. In the preliminary study, we screened out 3 differently expressed EBV miRNAs from EBV-induced lymphoma model by using miRNA chip. We found that ebv-miR-BART16 maybe modulate apoptotic signaling pathway of EBV-infected cells, which could be one of the mechanisms of EBV in inducing malignant transformation of lymphocyte. This project plans to validate the target genes of ebv-miR-BART16 related to apoptosis signaling pathway by using reporter gene technology. The malignant phenotype of lymphoma cell such as characters of anti-apoptosis and proliferation were detected by using tumor bearing mice model and lymphoma cell lines which is stably transfected with ebv-miR-BART16 ; and then we are trying to obtain the direct evidences about ebv-miR-BART16 modulating proliferation and apoptosis signaling molecules by detecting cell apoptosis and the molecular activity levels associated with apoptosis signal pathways and finally reveal the biological function of ebv-miR-BART16 and its role in lymphomagenesis.
