中文摘要: 随着纳米氧化锌(ZnO)的工业化生产,纳米ZnO释放进入环境发生人体暴露的安全性问题越来越受到关注。而纳米ZnO呼吸暴露的安全性研究大多受限于现有的研究手段,并不能真实反映人在真实环境下的暴露情况。我们将针对纳米ZnO呼吸暴露,创新性利用细胞水平的气液界面暴露装置和动物呼吸暴露实验,研究纳米ZnO产生的呼吸毒性效应,重点阐述纳米ZnO在气溶胶暴露状态下导致氧化应激、炎症反应、遗传毒性进而导致毒性损伤的机理。预期结果获得纳米ZnO不同物理化学性状与毒性效应之间的关系,阐明纳米ZnO产生相关毒性损伤的全面机制理论。本研究将为纳米ZnO生产现场暴露提供剂量效应阈值参考,为完善纳米材料生物安全性的评估提供参考方法,同时也为今后相关标准及法规的制定奠定必要的理论和实践基础。

英文摘要: With the rapid development of nanotechnology, an increasing attention has been focused on the nano-safety of nanomaterials. Nano-ZnO, as a new type of functional nanomaterials and having industrial level production, may generate adverse response to human health. Herein, we focus on the research of the inhalation toxicity of nano-ZnO by exploring its oxidative stress, inflammation, genetic toxicity and related toxic effects at the cellular and animal levels. The air-liquid interace exposure and inhalation exposure tests will be adopted to evaluate the associated endpoints of nano-ZnO inhalation toxicity. The implementation of this project will promote the study of the health effects of nano-ZnO in China, and produce reliable reference data for relevant environmental control threshold. Finally, this research will lay a substantial foundation on which the evalutaiton standards and regulations relevant to the nano-safety would be established.
