中文摘要: 氮素是农作物产量的重要限制因子。NRT1/PTR FAMILY(NPF)是与氮素(NO3-、寡肽等)吸收利用过程密切相关的一类转运蛋白。申请人从小麦中分离得到34个NPF同源基因,发现其中一个成员TaNPF8.1(B)的表达在小麦根中受NO3-诱导显著提高。本项目拟通过蟾卵异源表达系统分析TaNPF8.1(B)在不同浓度NO3-和pH条件下参与的NO3-转运功能,以及对其它底物如寡肽的转运功能;利用小麦原生质体确定TaNPF8.1(B)的亚细胞定位;通过过量表达TaNPF8.1(B)研究其在氮素吸收、转运中的生理功能,以及对生长后期氮素再分配过程的影响,为小麦中研究提高氮素利用率提供理论依据。

英文摘要: Nitrogen is a serious limiting factor of the yields of crops. NRT1 PTR FAMILY (NPF) genes encode transporters that are closely related with the nitrogen (primarily nitrate, oligopeptide etc.) uptake, long distance transport, distribution, and the regulation of nitrate uptake. Wheat (Triticum aestivum) is one of the most important crops, whereas research work of NPF genes in wheat is still absence. I have isolated 34 wheat NPF genes, and found that the expression of TaNPF8.1(B) was significantly induced by the nitrate. In this project, we’ll analyze the subcellular localization, heterologous expression in Xenopus Lavies oocyte to identify the function of the TaNPF8.1(B) as a plasma membrane-localized nitrate transporter. Further analysis of physiological function in nitrate uptake, long distance transport, and the influences on the nitrogen redistribution during reproductive growth stage will be in progress with the creation of wheat TaNPF8.1(B) overexpressing plants. This project is expected to provide theoretical basis for further research in order to improve the nitrogen use efficiency in wheat.
