中文摘要: 精神分裂症(SZ)是一种遗传度较高的重性精神疾病,基因调控异常可能是发病的关键环节。microRNA(miRNA)是一类重要的转录后负性调控因子,研究提示miRNA调控异常可能与SZ发病有关。研究发现外周血RNA分子反映了精神疾病在外周的特异变化且与中枢具有一定的相关性。本项目组的基因组表达谱芯片检测发现在首发SZ患者外周血中部分miRNA表达下调,而部分mRNA表达增高,生物信息学预测发现两者之间存在miRNA异常调控网络。本研究提出如下假设:miRNA介导的基因组转录后调控异常可能在SZ发病中发挥重要作用。本项目组拟利用病例-对照样本验证并筛选异常调控网络中miRNA和mRNA的表达水平,再利用分子实验对筛选出来的miRNA-mRNA结合关系进行生物学验证,最后对验证后的miRNA-mRNA调控网络进行通路分析。本研究预期结果有望为SZ发病机理研究提供实验证据。

英文摘要: Schizophrenia (SZ) is a severe mental disorder with a high heritability. Current studies indicate genetic dysregulation may play vital roles in the development of SZ. miRNA is a critical class of posttranscriptional negative regulators that may play a vital role in the development of SZ.It has been suggested that peripheral blood RNA levels may reflect a distinctmolecular response of the peripheral cells and may be correlated with contral nervous system. We found the predominant downexpression of miRNAs and predominant upexpression of mRNAs in SZ compared with healthy controls, and in silico prediction suggests the existence of abnormal miRNA regulation network between them. Thus, we hypothesize thatposttranscriptional genomic dysregulation mediated by miRNA may play a vital role in the development and treatment of SZ.The study seek to clinically verify these abnormal miRNA regulatory networks using case-control samples, to biologically verify the miRNA-target bindings using molecular experiments, and to conduct pathway analysis on the miRNA-mRNA regulatory network. The study may provide scientific data for the mechanism of SZ.
