中文摘要: 植物的生长发育调控机制复杂,涉及多条代谢和信号通路。脂质是生物体物质代谢、能量转换和信号转导的重要参与者,往往调控多条通路,广泛参与各种生物活动过程。磷脂酸磷酸酶(phosphatidic acid phosphatase, LPP)通过催化磷脂酸(PA)去磷酸化而产生二脂酰甘油(DAG),是调控脂质合成代谢途径的重要分支点。然而,植物LPP的生物学功能和分子机理尚未清楚。我们前期研究发现水稻磷脂酸磷酸酶OsLPPa5在植物生长发育中具有重要作用。本研究将进一步通过分子遗传、生化、代谢组学、细胞超微结构、Co-IP、蛋白组学和高通量质谱技术等方面系统阐明OsLPPa5的生物学功能,解析OsLPPa5介导的膜脂代谢在植物生长发育中的分子调控网络及其关键节点,填补膜脂代谢和信号的未知科学理论知识,并挖掘调控作物生长发育和产量的重要新基因,为作物高产育种提供理论依据和原创性成果。

英文摘要: Plant growth and development are regulated by multiple pathways, and the molecular mechanism is complicated. Lipids are vital players involved in various biological processes including metabolism,energy storage and signal transduction in cells. Phosphatidic acid phosphatase (LPP)catalyzes a crucial step in lipid metabolism via dephophorylating phosphatidic acid (PA) to produce diacylglycerol(DAG). Both PA and DAG are important regulators and signal messengers. However, the biological function and molecular mechanism of plant LPP are still unclear. Recently, we isolated a LPP gene from rice genome and found that LPP plays an important role in plant growth and development. To further characterize the biological function and regulation of LPP, we will perform systemic approaches including genetics, biochemistry, metabolism, and molecular interaction assay and mass spectrometry to dissect the signaling network mediated by LPP. The completion of this study will shed light on lipid metabolism and molecular mechanism in plant growth and development. It also will provide the valuable basis for improving crop plant production.
