中文摘要: 生物物理领域的一个重要发现就是,DNA不仅是基因存储和转录的基本材料单元,同样也是各种生物反应的催化剂。DNA本身不含有金属离子,但它对相当多的金属离子具有选择性亲和作用。DNA和金属的相互作用具有非常重要的意义。DNA的人工合成和筛选是目前合成生物学研究领域的热点之一,而金属离子作为辅基,在辅助DNA折叠、稳定DNA构象、参与活性位点构成等方面有重要作用。同时,利用DNA对不同金属的亲和作用,可以发展简单高效的金属生物探测器。不论是金属涉及的DNA催化剂,还是DNA金属生物探测器,它们的进一步发展都迫切需要我们从电子/分子水平上理解DNA-金属的相互作用机制。本项目在我们目前的金属蛋白质XAS研究基础上,拟通过XAS结合从头计算研究DNA与金属相互作用的局域三维精细结构。本项目的展开不仅拓展了目前BioXAS的研究领域,也为理解DNA-金属复合体系的相互作用提供电子/分子结构信息。

英文摘要: In the last decades, one of the most important discoveries is that DNA can act as enzymes, termed DNAzyme, as well as genetic materials. Although there are no metal ions in DNA, they have a selective affinity to few divalent ions. Most DNAzymes require metal ions for catalytic function, either through facilitation of the folding of DNA into stable tertiary structures, or through direct participation in the chemical reaction. And some biosensors can be designed against the selective affinity. Identification of the nature of interactions between DNA and metal ions is an important outstanding question for both DNAzymes and DNA biosensors. To better understand the molecular details of these interactions, it is essential that we have a reliable picture of the metal ion coordination site. This program would combine XAS and ab initio calculations to resolve and characterize the 3D fine structure of metal binding sties. As well, this program broadens the application sphere of BioXAS, and implies electron/molecular structure information of DNA-metal complexes.
