中文摘要: 膀胱移行细胞癌是我国泌尿系统最常见的恶性肿瘤,寻找其治疗的新方法非常重要。2006 年发现了小分子RNA 激活基因表达(RNAa)的作用(PNAS,103:17337)。此后该发现者与我们课题组合作,开展筛选靶向于膀胱癌抑癌基因启动子区的功能性saRNAs(小激活RNAs)的研究。在此基础上,本项目将采用FISH、基因芯片及高通量和定性定量的RNA 组学检测技术,深入分析saRNAs 作用的亚细胞定位、靶向特异性及其与微小分子RNA(miRNA)的关系,探明saRNAs 的若干作用机制;通过生物信息学结合分子生物学技术,进一步探索高效saRNAs 的序列特征。本项目的开展将对从新的层次阐明RNAs激活技术具有重要意义。

英文摘要: It's very urgent and important to develop new therapeutics because bladder transitionalcell carcinoma is the most common urological neoplasms in our country. Our cooperator,Li found the phenomenon of gene activation by small RNA in 2006 (PNAS, 103: 17337), and then cooperated with us to search the functional saRNAs which target the promoters of tumor suppresive genes in bladder cancer. To further research on mechanisms of saRNAs, we will deeply investigate the subcellular location, target specificity and relationship with miRNA of these saRNAs by FISH, genetic microarray, high throughput qualitative and quantitative detective technologies of RNA analysis. We will also investigate the characteristics of the sequences of effective saRNAs by combination of bioinformatics and molecular biology technologies. This project will be important for interpretation of the RNAs activation technology.
