中文摘要: 抗微管药物是治疗恶性肿瘤的一类化疗药物。崖豆藤属(Millettia)植物是我国民间广泛使用的草药。该属植物中富含结构丰富的黄酮、异黄酮、香豆素类、生物碱、三萜等类型的化合物,具有良好的抗肿瘤活性。前期研究表明本属植物中含有良好抗微管活性的天然活性成分。本项目拟通过对原产我国云南的十种崖豆藤属植物进行系统深入的研究,建立新型高效的分离分析方法,发现一系列新的和新骨架化学成分;构建该属植物的化合物的样品库和数据库;筛选新的抗微管治疗药物的化学实体,结合结构生物学,通过高分辨晶体结构指导基于微管蛋白α亚基的结构修饰,探讨构-效关系,发现和创制基于新靶点的全新抗微管、抗耐药的肿瘤治疗先导化合物,为发展新型抗肿瘤药物及其临床应用奠定试验基础。

英文摘要: Anti-microtubule drugs are an important type of cancer drugs. Plants in the genus Millettia are widely used in Chinese folk medicine and rich in flavonoids, isoflavones, coumarins, alkaloids, triterpenoids and other types of compounds. Previous studies have proved that the natural constituents isolated from genus Millettia plants had a good anti-microtubule activity. In order to discover a series of novel compounds with new skeleton and establish the sample libraries and databases, this project will study ten Yunnan Millettia genus systematically through new and efficient separation and analysis methods established, screen new anti-microtubule chemical entities and discuss the SAR based on the chemicals’ binding ability with high-resolution crystal structure of tubulin α subunit. In this way, novel anti-microtubule and anti-resistance lead compounds based on new target will be discovered, which lay a foundation for the development of cancer drug and its clinical applications.
