中文摘要: 药物代谢动力学参数的研究,对器官功能评估、疾病诊断、药物研发等有重要的意义。荧光分子断层成像技术具有灵敏度高、特异性强、成像快、无电离辐射损伤等优势,在基于小动物模型开展的生物医学研究中具有独特优势。本项目中拟研究建立荧光分子动态参数反演成像方法,以期通过生物体表的荧光检测信号,在解剖结构先验信息的引导和约束下,直接反演计算获取能够表征生物体内各组织器官固有生理功能特性的药代动力学参数图像,为基于药代动力学参数开展的生物医学研究提供有力工具。研究内容包括:(1)研究建立将药代动力学参数映射到体表荧光检测信号的理论模型,以期为基于体表检测信号直接反演计算获得参数信息提供理论基础。(2)研究建立基于结构先验的荧光分子动态参数直接反演成像算法,以期在结构先验信息引导约束下,直接反演计算获得各组织器官的参数图像。(3)开展仿真实验对算法进行测试和优化。(4)开展活体实验验证算法在活体应用中可行性。

英文摘要: The study of pharmacokinetic parameters plays an important role in organ function evaluation, disease diagnosis and drug development. With the advantages of high sensitivity, high specificity, fast and nonionizing radiation imaging, fluorescence molecular tomography has unique superiority in biomedical studies with small animals. In this project, pharmacokinetic parameter reconstruction method with structure a prior in fluorescence molecular tomography will be studied, in order to get pharmacokinetic parameter images which could represent the inherent unique physiological function of different tissues and organs from boundary fluorescence measurements. The method will provide a powerful tool for biomedical studies based on pharmacokinetic parameters. The research contents include: (1) The theoretical model which maps the pharmacokinetic parameters to the boundary fluorescence measurements will be studied and constructed, in order to provide a theoretical basis for the reconstruction of parametric images directly from the raw boundary measurements. (2) The pharmacokinetic parameter reconstruction algorithm with structure a prior will be studied and constructed, in order to reconstruct parametric images directly from the raw boundary measurements.
