中文摘要: 基质金属蛋白酶家族(MMPs)、成纤维细胞(FB)增殖活性、端粒长度等在皮肤光老化过程中发挥关键作用。研究表明,上述因子很可能也参与了强脉冲光(IPL)嫩肤过程。本研究采用长波紫外线(UVA)照射人皮肤和FB构建光老化模型,观察IPL照射前后上述因子的变化。结果发现:(1)正常皮肤经UVA反复照射后真皮弹力纤维碎裂变性;胶原排列稀疏,MMP-1,3,9,12,TIMP-1表达增加,成功构建了皮肤光老化模型。(2)与自然恢复组光老化皮肤相比,IPL照射组光老化皮肤胶原排列均匀致密,I型胶原表达增加,MMP-1,3,9,12,TIMP-1表达水平升高,MMP-9升高最为明显。(3)UVA联合8-甲氧补骨脂诱导的老化FB增殖活性下降,端粒长度缩短,活性氧增加,β#21322;乳糖苷酶及MMP-1水平升高。IPL照射可降低老化细胞内活性氧及MMP-1水平,阻止端粒缩短。(4)IPL可减少G1期FB比例,增加S期细胞,促进CyclinD1和CDK2的表达。以上结果表明IPL可从在体及细胞水平逆转UVA造成的光老化。

英文摘要: Matrix metalloproteinase, fibroblast cell viability and telomere length play critical role in skin photoaging. Investigations suggest these factors may involve in the process of IPL photorejuvenaton. In this study, UVA induced photoaged skin and fibroblasts were irradiated by IPL and the expression of these factors was examined. The results: (1) Compared with the normal control skin, there were fragment of elastic fibers, sparse collagen fibers and significant elevation of MMP-1, -3, -9, -12, TIMP-1 levels in UVA induced photoaged skin. Photoaged skin model has been successfully constructed. (2) Compared with the spontaneously recovered photoaged skin, there were elevated collagen I mRNA levels and more compact arrangement of collagen fibers and significant elevation of MMP-1, -3, -9, -12, TIMP-1 at protein and mRNA levels in IPL irradiated photoaged skin. (3) There were decreased cell viability, shortened length of telomere, higher level of ROS, increased βalactosidase and MMP-1 expression in PUVA induced photoaged fibroblasts. Irradiation with IPL after PUVA exposure prevented telomere shortening and decreased the ROS and MMP-1 levels in comparison with PUVA treatment. (4) IPL decreased the G1% of cell cycle, increased the S% of cell cycle, and promoted the expression of CyclinD1 and CDK2 in fibroblasts. These results demonstrated that IPL could partially attenuate the UVA-induced photoaged skin in vivo and fibroblasts in vitro. The different effects of IPL on human fibroblasts and skin tissue may result from the different dose, frequency of IPL irradiation and the difference between cell and tissue, which contribute to the difference in MMP expression.
