中文摘要: microRNA对细胞的基因表达、发育分化具有重要的调控作用,是生命科学领域研究的热点。实现microRNA 的无标记检测对于癌症诊断和治疗有着十分重要的应用价值。而最理想的无标记检测手段应该具有敏感度高、选择性好、易集成化、便携性好甚至是低成本可抛的特点。针对此,本项目将研究基于溶液法低电压有机薄膜晶体管(organic thin-film transistor, OTFT)的microRNA传感器件。结合对microRNA分子生物学特性.的研究,理清检测灵敏度与OTFT器件电学特性之间的内在关系,建立器件性能优化设计和工艺调控的方法,设计与实现集成式的传感器件用于microRNA分子的检测,为发展高灵敏度、低成本、可抛式的microRNA检测手段打下基础。

英文摘要: The regulation of microRNA on the gene expression, development and.differentiation is currently a hot research topic in biological science. It.is of great value for cancer diagnosis and treatment to achieve label-free microRNA detecting. However, the ideal label-free detecting way should be of high sensitivity and excellent selectivity, easy to be integrated, portable and even low cost and disposable. Therefore, this project proposes to develop a type of microRNA sensors based on the solution processed low voltage organic thin-film transistor (OTFT). With the study of the biological properties of the microRNA molecules, this project will investigate and understand the mechanisms of how the OTFT electrical properties affecting the sensitivity of the sensor devices, then develop the methods for device performance optimization and process control, and finally design and fabricate an integrated sensor structure for microRNA detecting.This project is expected to form an important basis for achieving a high sensitivity, low cost and disposable microRNA detecting method.
