中文摘要: 锕系元素均是有毒的放射性重金属元素,随着核能事业的快速发展,人们越来越关注核燃料循环过程中释放出来的锕系元素对环境和生物体的影响。研究锕系元素阳离子与生物分子尤其是蛋白质或多肽的相互作用机理,对于理解锕系元素阳离子在生物体内的吸收与转运机制至关重要,对设计基于多肽复合物的锕系元素促排解毒药物亦具有重要意义。本项目拟以有重要生理意义的多肽分子为原型,通过分子设计,合成出能选择性结合锕系元素(主要研究U、Pu)阳离子的多肽分子和磷酸化多肽复合物。利用多种手段如紫外可见光谱、X-射线精细吸收谱、荧光光谱、质谱、圆二色谱、电位滴定、红外光谱等研究磷肽与锕系元素阳离子的作用机制,尤其是配合物结构和配位模式,并研究磷酸化修饰对锕系元素(U、Pu)阳离子配位模式的影响。为深入理解锕系元素阳离子与蛋白质的结合机理和设计基于活性多肽复合物的促排药物奠定理论与实验基础。

英文摘要: Actinides are toxic and radioactive heavy metals. With the rapid development of nuclear energy industry, more and more attention has been paid to the radioactive actinides which can be released into the environment by nuclear fuel cycle activities. Investigating the bio-toxicology of actinide cations and their interaction mechanisms with biomolecules is of great significance for evaluating the harzard of biological exposure to actinides, and also for designing high efficient peptide-based actinide chelating drugs for detoxification. In this project, peptides with important physiological significance are initially selected as models. After specific molecular design, peptides and phosphopeptide complexes with selective affinity with actinides (mainly U and Pu) will be synthesized. The interaction mechanisms between actinide cations and peptide complexs will be systematically studied by different spectroscopic methods such as NMR, UV-Vis, Infrared, Fluorecence, Circular Dichroism, Mass spectrum and X-ray absorption fine structure spectrum, especially concentrating on the coordination structures of the complexes. The effect of peptide phosphorylation on the coordination will be ravelled.
