中文摘要: 肥胖和2 型糖尿病对我国公共卫生医疗造成了很大负担,其发生和发展机制的研究是一项巨大的挑战。肥胖和2型糖尿病是由环境因素和遗传因素共同作用的复杂代谢疾病,因此,表观遗传学效应,尤其DNA甲基化,在其发生和发展过程中可能扮演很重要的角色。本研究以高脂诱导肥胖和糖尿病模型小鼠为研究材料,运用MeDIP测序和MSCC测序两种全基因组甲基化测序技术,研究在饮食环境因素的影响下,肥胖和二型糖尿病发生、发展过程中DNA甲基化的改变,从而更全面的解释糖尿病的病例机制。本研究还将在人群中验证甲基化改变的差异基因,分析肥胖或糖尿病相关的甲基化差异基因在人群中的作用效应,为糖尿病的预防、治疗等提供医学转化的理论依据。

英文摘要: Obesity and type 2 diabetes have caused great burden on our public health, and the study of mechanism on their occurrence and development is still a great challenge. Obesity and type 2 diabetes are complex metabolic diseases, which affected by both environmental and genetic factors. Therefore, epigenetic effects, especially DNA methylation, may play very important role in the occurrence and development processe. In this study, for more comprehensive explanation of the mechanism of diabetes, we use high-fat diet induced obese and diabetic model mice as experimental material, and use two genome-wide methylation sequencing technology (MeDIP sequencing and MSCC sequencing) to investigate the changes of DNA methylation in the process of obesity and diabetes, which is under the influence of high-fat diet. In addition, this study will be also to confirm the changes of methylation in a special population, and to evaluate the effects of obesity or diabetes associated differentially methylated genes, so we can provide the theatric basis on medical transformation in the prevention and treatment of daibetes.
