中文摘要: 作为各种疾病重要的诊疗新靶标,血液和唾液中的微泡在疾病的发生和发展过程中发挥着重要作用。本项目通过发展新方法,实现对血液和唾液中微泡的定量蛋白质组学分析,并应用到肺癌的诊疗中。具体研究目的是:1、建立血液和唾液微泡的分离制备新方法;2、建立微泡的蛋白质组学研究新技术和新方法;3、揭示血液和唾液中微泡的蛋白质组成与差异;4、在微泡中发现肺癌的特异性蛋白质生物标志物;5、发展新的技术和方法,对微泡中肺癌相关的蛋白质生物标志物进行定量分析,对选定的生物标志物进行初始验证和独立验证。研究意义在于:1、确立微泡的分离制备新方法,为微泡的临床应用奠定基础;2、阐明血液和唾液中微泡的蛋白质组成和差异;3、在微泡中发现肺癌和慢性阻塞性肺病的特异性蛋白质生物标志物,为肺癌的诊疗提供生物学依据;4、实现微泡中肺癌生物标志物的特异性检测和评价,为肺癌的无损伤检测提供分析手段。

英文摘要: Microvesicles in human blood and saliva have been visioned as new therapeutic targets in the clinic, which play key roles in diseases' progression, metastasis and angiogenesis. Through developing new methods, the aim of this proposal is to quantitatively analysis the proteome in the microvesicles of lung cancer patients. The goals are as follows. 1. Establish the new methods for microvesicles preparation in human blood and saliva; 2. Establish the new proteomic technologies and methods for the analysis of microvesicles proteome; 3. Unveil the proteomic difference of microvesicles from human blood and saliva; 4. Discover the proteomic biomarkers in the microvesicles of blood and saliva for lung cancer; 5. With the help of developing new technologies and methods, detect the potential biomarkers in microvesicles for diseases quantitatively. The significance of this research is as follows. 1. The developed simple and efficient new methods for microvesicles preparation will contribute to its clinical applications; 2. Clarify the protein composition and difference between blood microvesicles and saliva microvesicles; 3. Discover valuable proteomic biomarkers in microvesicles for the clinical diagnosis of lung cancer and chronic obstructive pulmonary disease; 4.
