中文摘要: 翘嘴鳜是我国特有的淡水经济鱼类,然而种质资源的退化严重制约着翘嘴鳜养殖业的可持续发展,亟需持久地对翘嘴鳜重要经济性状进行遗传改良,因此积极开展翘嘴鳜分子标记辅助育种研究具有重要的理论与应用意义。本项目以生长性状为切入点,通过翘嘴鳜高密度遗传连锁图谱构建和生长性状QTL多家系定位分析,可有效地为分子标记辅助育种提供可靠性高的分子标记,内容包括:(1)在一个杂交F1家系中利用SSR和SNP构建翘嘴鳜高密度遗传连锁图谱并进行生长性状QTL精细定位;(2)利用定位在图谱上的SSR标记在另外两个杂交F1家系中进行生长性状QTL初步定位,并利用位于QTL置信区间内的SNP标记(序列延伸)对QTL作进一步定位;(3)通过QTL比较分析,有效筛选与生长性状紧密连锁且可靠性高的分子标记。本项目的完成不仅为翘嘴鳜基因组学研究奠定重要基础,也为其分子标记辅助育种体系建立和优良品种培育提供理论依据和技术方法。

英文摘要: Chinese perch (Siniperca chuatsi) is a valuable freshwater fish restricted to China, however, the degradation of germplasm resources has seriously impeded its sustainable development of industrialization aquaculture. It is urgent to persistently carry out genetic improvement for important economical traits in S. chuatsi, therefore, it is of important meaning in both theory and application to actively carry out research on molecular marker assisted breeding. The project uses growth traits as a breakthrough point to effectively provide high reliability molecular markers for molecular marker assisted breeding through high-density genetic linkage map construction and QTL mapping for growth traits in multi-families, including: (1) constructing the high-density genetic linkage map for S. chuatsi with SSRs and SNPs and QTL fine mapping for growth traits in a F1 hybrid family; (2) QTL mapping using SSRs located on the high-density genetic linkage map and further QTL mapping using SNPs (sequences extension) located in QTL confidence intervals for growth traits in another two F1 hybrid families; (3) screening high reliability molecular markers closely linked to growth traits through comparative QTL analysis. The accomplishment of the project would not only lay important foundation for genomics research, but also provide theoretical basis and technical method for molecular marker assisted breeding system establishment and excellent strain breeding in S. chuatsi.
