中文摘要: 本项目旨在研究釉原蛋白(AML)诱导系统骨髓间充质干细胞(BMMSCs)归巢在牙周组织缺损治疗中的作用及其机制。基于国际上对系统骨髓干细胞在牙周组织代谢平衡中所起作用研究的最新进展,以及本项目组前期对AML促进BMMSCs迁移的研究结果,本项目拟证实AML通过Wnt/β-catenin信号通路促进其迁移,并采用绿色荧光蛋白小鼠骨髓移植结合急性牙周缺损模型,观察AML及β-catenin激动剂Wnt3a作为AML的小分子替代物在诱导BMMSCs向牙周组织归巢并参与牙周组织再生过程中的作用。在此基础上,进一步探讨将Wnt3a应用于牙周缺损治疗的可行性。目前国内外从诱导系统BMMSCs归巢的角度促进牙周再生的研究还未见报道。本项目的研究结果可为牙周再生研究提供新的思路,为牙周组织缺损的临床治疗提供新的药物和方法,也可为种植义齿种植体周围的牙周再生以及牙齿再生研究提供理论和技术依据。

英文摘要: The aim of this study is to evaluate the role and mechanism of amelogenin in the treatment of periodental defects by inducing systemical bone marrow mesenchymal stem cells(BMMSCs)' homing. Based on the latest progress about the role of bone marrow-derived mesenchymal stem cells in the maintenance and regeneration of periodental tissue and our preliminary data suggesting that AML promotes the migration of BMMSCs, our proposed experiments will 1)use the chamber system to demonstrate that AML promotes the migration of BMMSCs by activing Wnt/β-catenin pathway.2)use the GFP+ bone marrow chimeric combined with periodental defects mice model to observe the role of AML and Wnt3a,the small molecule substitute, in inducing BMMSCs'homing to the periodental tissue and promoting the periodental regeneration.3)study the feasibility of using Wnt3a for periodental regeneration.The study on promoting periodental regeneration by enhancing systemical BMMSCs'homing has not been reported. Our findings will provide new strategy for the study of periodental regeneration,new medicine for the treatment of periodental defects, new theoretical and technical basis for periodental regeneration around implants,as well as tooth regeneration.
