中文摘要: 肾保护因子在急性肾损伤(AKI)的修复中起重要作用。Persephin是与肾脏发育密切相关的因子,研究表明PSP可以通过激活MAPK途径发挥增殖及抗凋亡作用,其对AKI的影响目前尚无报导。我们前期研究在证明人早胚间充质干细胞(hEMSC)对AKI具有修复作用的基础上,采用抗体芯片分析发现hEMSC分泌其他干细胞未报导的细胞因子PSP,进一步观察到PSP对体外损伤肾小管细胞也有修复作用,我们推测PSP对于AKI有修复作用。本研究拟在顺铂诱导小鼠急性肾损伤模型中探讨PSP的肾保护作用,主要针对PSP对小管细胞的增殖及凋亡作用等关键环节进行研究;并在体外试验进一步验证其发挥作用的受体及信号通路。本研究将为AKI的防治提供新的备选肾保护因子,为以细胞因子为靶点探寻更特异有效的防治KAI的发展恶化手段提供新思路和方向。

英文摘要: The renal protective factors play an important role in the repair of acute kidney injury (AKI). Persephin closely related with kidney development, studies show PSP can increase proliferation and decrease apoptosis by activating the MAPK pathway. Currently, the role of PSP in AKI is not be reported. In preliminary studies, we have proved that the early embryo-derived mesenchymal stem cells (hEMSC) ameliorate AKI. Furthermore, using antibody microarray analysis, we found that hEMSC secret PSP which has never been previously unreported. In view of the PSP has an indispensable role in kidney development process, the pre-experimental observations of PSP having therapeutic effect on injured renal tubular cells, we hypothesized that PSP plays an renoprotective role in AKI. The PSP renoprotective function is futher proved by AKI animal models and injured renal tubular epithelial cell models. In vitro, cultured cell model is used to testify the receptor and activated signaling channels of PSP.
