中文摘要: 弧菌是对虾的主要细菌性病原。鞭毛蛋白(Flagellin)是弧菌重要的毒力因子,但对虾如何感知鞭毛蛋白尚未有研究。我们发现日本囊对虾中一种新型模式识别受体Leurelectin可通过其含有的富亮氨酸重复区(Leucine-rich repeats)识别鳗弧菌鞭毛蛋白 A(FlaA)。在此基础上,本项目拟检测Leurelectin对多种病原弧菌鞭毛蛋白的结合作用,确定其识别范围;研究Leurelectin与FlaA结合的作用机制,发现决定识别的关键部位;干扰和过表达Leurelectin,确定其在FlaA诱发的免疫反应中的具体功能;构建鳗弧菌FlaA突变菌株,进一步明确Leurelectin-FlaA结合在应对鳗弧菌感染中的作用;鉴定Leurelectin的下游互作蛋白,揭示其参与的免疫信号转导途径。本研究将首次发现无脊椎动物中的鞭毛蛋白受体并阐述其功能,可为对虾细菌性病原防治提供新思路。

英文摘要: Vibrio is the main bacterial pathogen of shrimp. Flagellin is the key virulence factor of Vibrio. However, how shrimp sense flagellin is unknown. We found a new pattern recognition receptor Leurelectin, which contained several Leucine-rich repeats, could recognize the flagellin A (FlaA) from Vibrio anguillarum. In this study, we will: check whether Leurelectin can bind other kinds of Vibrio flagellins to confirm its binding spectrum; study the Leurelectin-FlaA interaction to reveal the critical sites for binding; suppress or over-express Leurelectin to discover its function in the FlaA-induced immue response; generate the FlaA-mutant to further demonstrate the function of Leurelctin-FlaA interaction to resist Vibrio anguillarum; identify the interacting protein downstream of Leurelectin to find out the immune signal pathway it is involved in. This study will firstly report a flagellin-receptor and its function in invertebrates, and also provide new insight to prevent and control the shrimp bacterial pathogen.
