中文摘要: 开展新颖纳米材料在重大疾病尤其是癌症的检测、诊断及治疗方面的研究越来越受到人们的关注,并成为近年来的研究热点之一。本研究以两嵌段共聚物聚苯乙烯-b-聚丙烯酸(PS-b-PAA)和Fe3O4纳米粒子为研究对象,在表面活性剂十六烷基三甲基溴化铵(CTAB)的结构导向下,利用自组装工艺和溶胶-凝胶方法,构建一种具有双介孔(小孔壳和大孔核)孔道、孔径可调以及磁性能可控的核壳型超顺磁性双介孔氧化硅纳米结构,并通过探索双介孔结构与磁共振成像性能之间的关系,为进一步设计制备性能优异的氧化铁基磁共振对比剂提供新思路和理论指导。然后采用浸渍法分别将两种不同性质的抗癌药物分子(亲水的盐酸阿霉素和憎水的紫杉醇)担载到双介孔结构的小孔和大孔孔道中,探索其在磁共振成像引导下癌症治疗方面的初步应用研究。本项目的意义在于为制备一类集高效MR诊断与治疗功能于一体的新型纳米诊疗剂提供新的方法和思路,具有一定的科学研究价值。

英文摘要: Recently, novel nano-materials have attracted more and more attention due to their potentials in detection, diagnosis and treatment of diseases, especially for cancer. In this proposal, core-shell structured superparamagnetic dual-mesoporous silica nanocomposites consist of magnetite-incorporated core with larger pores and shell with smaller pores will be designed and synthesized by employing the diblock copolymer of polystyrene-b-poly (acrylic acid) (PS-b-PAA), Fe3O4 nanoparticles and the surfactant cetyl trimethyl ammonium bromide (CTAB) based on the self-assembly and sol-gel techniques. The pore size and magnetic properties will be tuned by controlling the experimental parameters. The relationship between the structure and the performance of magnetic resonance imaging will be investigated. These could provide new ideas and theoretical guidance for the design of the excellent iron oxide-based magnetic resonance contrast agent. On the basis of the above research, two different kinds of anti-cancer drug molecules (hydrophilic doxorubicin hydrochloride and hydrophobic paclitaxel) will be loaded into the pore channels using the impregnation method, respectively, so as to explore the magnetic resonance imaging-guided cancer therapy effect.
