中文摘要: 乳腺癌是我国最常见女性癌症,且正以世界第一的增速(每年3%)高速增长,是我国亟待解决的重要健康问题。超过90%乳腺癌患者的死亡原因是由于肿瘤迁移,目前迁移性乳腺癌患者的五年生存率尚不足20%,且无任何有效治疗药物。因此,我们提出了一种可以精确靶向迁移性乳腺肿瘤的个体化抗癌药物。我们首次将“多元协同靶向”技术用于个体化抗癌药物研究:通过表征10名迁移性乳腺癌患者的2种特异靶点(ICAM-1和EGFR),建立起每位患者的个体化靶点组合,用于修饰脂质体药物模型。所得到个体化“多元协同靶向”药物通过与患者癌细胞膜脂筏中的ICAM-1和EGFR表达进行互补,既能提高靶向药物对肿瘤细胞靶向效率,精确的将药物运输到肿瘤位点,减少对正常器官的副作用;又能对癌细胞的ICAM-1和EGFR信号通路进行协同阻断,达到同时抑制肿瘤生长和迁移的协同治疗效果,为研究个体化抗癌药物提供了新理论和治疗技术。

英文摘要: Breast cancer is a major global health issue, and currently the most common form of women cancers worldwide. The primary cause of morbidity in these patients was tumor aggressiveness, characterized by metastases to regional lymph nodes, bone marrow, lung and liver. These sobering statistics, coupled with the fact that 20% of women diagnosed with breast cancer have advanced disease at the time of intial presentation, highlight the significant and unmet need for the development of therapeutics that treat aggressive and metastatic breast cancer (MBC). In this proposal, unlike conventional therapeutics that present a single targeting ligand homogeneously on a nanoparticle surface, we propose to engineer dual-targeting liposomal vehicles that complement the relative molecular density based on individual patients' pathological condition. This proposal will develop a therapeutic designed to precisely target mechanisms that govern MBC proliferation and metastasis, which may provide a significant opportunity to extend MBC patient survival.
