中文摘要: 本项目完全按照计划执行。首先,分别从当归、枸杞、党参、银耳、黄精、牛膝、麦冬、大枣、黑木耳、白术、山药、大蒜、板蓝根等13种中药中提取总多糖和分级多糖,比较了它们的抗病毒和增强免疫活性,筛选出各多糖的活性部位。第二,用正交试验或响应面试验优化了以上13种多糖的硫酸化修饰条件,建立了各多糖的硫酸化修饰方法,对筛选出的各多糖活性部位纯化后进行硫酸化修饰。第三,比较了以上13种硫酸化多糖(每种3~9个硫酸化多糖)的体外和前9种硫酸化多糖(每种2~6个硫酸化多糖)的体内抗病毒和增强免疫活性,分别筛选出各多糖活性最强的硫酸化多糖,并研究了其作用机理。第四,从前期研究和本项目研究筛选的16种硫酸化多糖中选择活性较强的10种硫酸化多糖进行总体比较,筛选出活性最强的硫酸化多糖;以此组成多个复方,通过试验比较筛选出1个增强免疫作用最好的方剂,命名为芪藿糖増免剂,并按新兽药研制要求完成了安全性、有效性、质量可控性研究,申报了新兽药注册。

英文摘要: This project has been performed fully according to the proposal. Firstly, the total and fractional polysaccharides were extracted respectively from 13 kinds of Chinese herbal medicines, Chinese angelica, Lycium barbarum, Codonopsis pilosula, Tremella, Siberian solomonseal rhizome, Achyranthes root, Ophiopogon root, Jujube, Auricularia aurlcula, Atractylodes macrocephala, Common yam rhizome, Garlic and Isatis root. Their antiviral and immune-enhancing activities were compared and the active sites for each polysaccharide were picked out. Secondly, the sulfated modification conditions of above-mentioned 13 kinds of polysaccharides were optimized by orthogonal or response-surface test. The sulfated modification methods for each polysaccharide were established and with which the active sites of 13 polysaccharides were sulfatedly modified after purification. Thirdly, the antiviral and immune-enhancing activities were compared of above-mentioned 13 kinds of sulfated polysaccharides (sPSs) (each including 3-9 sPSs) in vitro and front 9 kinds of sPSs (each including 2-6 sPSs) in vivo. The sPSs with the strongest activity for each kind of polysaccharide were picked out and their action mechanisms were claritied. Finally, the total comparison of 10 kinds of sPSs with the stronger activity from 16 kinds of sPSs selected by the research in early days and this project were performed. The sPSs with the strongest activity were picked out, with which several prescriptions were composed and a prescription with strongest immune-enhancing activity, namede Qihuotang immunopotentiator, was picked out by experiments. Its safety, effectivity and quality controllability were investigated according to developing request of new veterinary drugs. It had been applied for registration of new veterinary drug.
