中文摘要: 传统高压脉冲电场非热杀菌系统需要提供几千伏甚至更高的脉冲发生器,从而使设备成本提高,应用受限。本项目拟研制一种适于非热杀菌的柔性微芯片脉冲电场(microchip pulsed electric field,简称MPEF)技术。借助 COMSOL 软件仿真分析电极结构,利用聚酰亚胺基底加工制成柔性芯片,从而搭建脉冲电场微系统实验平台。以蓝莓汁为处理介质,利用MPEF对微生物钝化的动力学进行分析,建立数学模型;通过对MPEF作用后微生物的亚致死、胞内蛋白质、胞内DNA、细胞膜、胞内酶和胞内mRNA变化研究,从分子水平上明确MPEF技术对微生物钝化的作用机理;采用香气活性值(OAV)和频率检测-气相色谱-嗅闻技术(FD-GC-O)研究MPEF非热处理后的蓝莓汁低温贮藏特征香气组分动态变化。上述研究结果可为MPEF更为广泛应用于包括蓝莓汁在内的果蔬汁的非热加工技术开展和应用提供理论支持。

英文摘要: The conventional PEF sterilization system need high-voltage pulse generator, which is difficult in manufacturing and too expensive for widespread applications. Therefore, our program will develop a kind of flexible microchip pulsed electric field (MPEF) technology suitable for non-thermal sterilization. COMSOL multiphysics software will be used to simulate the electric field distribution on the microchip; flexible polyimide (PI) film based microchips will be fabricated. Then a variety of parameters will be studied in the PEF experimental platform. Take blueberry juice as material, we'll do research about changing dynamics of this MPEF non-thermal processing new technology on passivation of microorganisms, and then build the mathematical model. Through the effects on the sublethal injury of microorganisms, intracellular protein, intracellular DNA, cell membrane, intracellular enzymes and the level of intracellular mRNA,we can understand the mechanism of microbial inactivation by MPEF technology on the molecular level. Finally , we will study the changes of various active ingredients of blueberry juice before and after MPEF non-thermal treatment through odor activity value(OAV) and frequency detection - gas chromatography- olfactometry(FD-GC-O). The above experiment can provide theoretical support for MPEF technology being more widely used in the development and application of non-thermal processing technology of varieties of fruit and vegetable juices, including blueberry juice.
