中文摘要: 随着荧光识别及检测技术的不断发展,对于一些重要的生物小分子,如硫醇小分子(Cys,Hcy,GSH),的荧光识别及成像研究逐渐成为近期的研究重点。目前,在基于硫醇小分子识别的荧光探针研究中,普遍存在荧光响应信号单一、结构相似化合物间干扰严重,不能实现水相环境检测等问题;对于这些生物小分子的荧光识别和成像研究也主要集中在可见光区和单细胞成像阶段。鉴于此,我们计划开发4-6种基于类氟硼吡咯荧光团检测硫醇小分子的水溶性比率型荧光传感器,实现专一性强,灵敏度高,水相工作,荧光信号比率变化的检测目标。同时计划开发3-5种基于类氟硼吡咯荧光团的近红外荧光发射检测硫醇小分子的荧光传感器。深入开展该系列荧光探针分子的生物学应用研究,在体外单细胞成像研究的基础上,实现生物活体体内荧光成像及与硫醇结合过程的动态成像监测,推动生物小分子荧光成像及荧光分析法的进一步发展。

英文摘要: With the development of fluorescent detecting techniques, the research of fluorescence sensing and fluorescence imaging for some important biology small moleculars, such as thiols (Cys, Hcy,GSH), has been an active research ares in recent years. For the fluorescence thiols sensors, the problems, such as unidirectional transmission, interference from the moleculars with similar structures, limits of the detection in the aqueous media, can not be ignored. Meanwhile, the fluorescence sensing is mainly in the visible region and fluorescence imaging is usually tested in the single-cell recording way. Therefore, we plane to develop 4-6 hydrosoluble fluorescent thiols sensors based on BODIPY analogues, which could work in the aqueous media in the ratiometric way with high sensitivity and high selectivity. 3-5 BODIPY analogues-based fluorescent thiols sensors, which could work in near infrared region, will be explored in the meanwhile. Based on the single-cell fluorescence recording, the exploration of the biology application of these fluorescence sensors to realize the records in vivo and the dynamic imaging is our goals. Through all these efforts, we wish we could promote the developments of the research of fluorescence sensing and fluorescence imaging.
