中文摘要: 阿尔茨海默氏病是最常见的慢性退行性疾病之一。β-淀粉样蛋白(Aβ)是脑内神经炎症的起始诱导分子,其介导的炎症反应对AD起始、进展速度和疾病严重程度具有重要的影响。大量体、内外研究表明大麻CB2 受体激动剂在Aβ诱导的炎症反应不同时期都具有抗炎作用,但对于炎症动态变化下CB2受体介导的脑内固有和适应性免疫调节作用仍不清楚。本课题采用CB2受体敲除小鼠动物模型,模拟AD病理模型,确证不同时间CB2受体在中枢固有免疫和适应性免疫反应中发挥免疫调节的关键作用,为大麻CB2受体成为治疗神经退行性疾病的神经源性炎症候选药物靶标提供有效的体内实验依据。

英文摘要: Alzheimer's disease (AD) is now one of the most common neurodegenerative diseases.Aβ oligomers would be responsible for the onset of neuroinflammation,accelerating the disease progression leading to severe pathogenesis.CB2 agonists exert the immunoregulatory functions in various inflammation state induced by Aβ in vivo and in vitro,but the direct role targeting to CB2 receptor remian undertermined.Our aim is to creat AD model base on CB2 receptor knockout mice and to identify the key role played between the innate and adaptive immune system in CNS by mimicing inflammation dynamic characteristics in aging condition with AD. Our research will provide much credible data in vivo that might explain the promising candidate drug target in treatment of neuroinflammation in neurodegenerative diseases.
