中文摘要: 三唑磷是一种具有潜在内分泌干扰效应的亲脂性有机磷农药,易在贝类性腺中积累,因此开展三唑磷生殖毒性研究是揭示其毒性机制和健康风险评价的必要前提。目前,有关三唑磷对海洋贝类生殖毒性研究尚未见报道。本项目针对我国海水养殖业中大量使用三唑磷,严重损害养殖生物与渔业资源的现状,开展三唑磷对翡翠贻贝的生殖毒性效应及其分子机制研究。通过观察三唑磷对翡翠贻贝产卵率、孵化率、性腺超微结构、细胞凋亡和激素水平的影响特征及其对性腺的氧化损伤作用,系统阐明三唑磷农药对翡翠贻贝的生殖毒性效应。通过SSH文库和代谢组学研究,获得三唑磷作用下贻贝性腺中差异表达基因和发生响应的小分子代谢物种类, 初步探讨三唑磷对翡翠贻贝生殖毒性的分子机制。本项目研究结果可为三唑磷农药对海洋生物生殖毒性效应及分子致毒机理的阐明提供理论基础,亦可为海水养殖业中施用三唑磷农药的生态风险评估及我国海水水质标准制定提供科学依据。

英文摘要: Triazophos, with a potential endocrine disrupting effect, is a lipophilic organophosphate pesticide, which displays higher bioaccumulation in the gonad of shellfishs. Therefore, research on its reproductive toxicity effect is necessary so as to elucidate the toxic mechanisms and health risk of triazophos. However, there is no report about the reproductive toxicity of triazophos in marine mollusks so far. Based on the extensive use of triazophos in our marine aquaculture and the subsequent threat to aquaculture and natural fishery resources, we investigate the reproductive toxicity and molecular mechanisms of triazophos on the green mussel Perna virudis. We investigate the triazophos influences on spawning and hatching rates, ultrastructure and cell apoptosis of gonad, steroid contents and antioxidant enzymes activity in order to systematically illustrate the reproductive toxicity of triazophos. By using the suppression subtractive hybridization method and NMR-based metabolomics, differentially expressed genes and the responded metabolites with low molecular weight in the gonad of the green mussel after treated with triazophos are indentified.The results of this proposal will lay a foundation for elucidating the reproductive toxicity effect and its molecular mechanism of triazophos in marine organisms, and also p
