中文摘要: 电化学DNA生物传感器是一种先进的基因检测技术,具有迅速、简单、成本低、灵敏度高、选择性好、可实现微型化等优点。本项目拟从材料设计角度出发,通过精确控制制备工艺获得不同结构的ZnO/金刚石电极,采用光化学和电化学方法对ZnO表面进行功能化修饰,利用电化学方法(循环伏安法、差示脉冲伏安法)和多种结构分析手段(SEM、TEM、XPS等)研究电极表面结构对薄膜表面功能化和传感器性能的影响,阐明电极表面结构与单链DNA探针的作用机制,探讨电极结构-表面功能化-传感器性能之间的内在联系,提出高性能电化学DNA传感器的设计原则,获得高灵敏度、高选择性的电化学DNA生物传感器。本项目研究成果不仅为研究生物分子与固体电极相互作用机制这一关键科学问题提供可靠的实验依据,更为构建高性能生物传感器提供设计思路和技术方法,为电化学DNA生物传感器朝向商业化发展奠定理论和技术基础。

英文摘要: Electrochemical DNA biosensor is one of novel techniques for the detection of DNA sequences, which has received wide attention due to its quick response, simple operation, low-cost, high sensitivity, high specificity and availability of miniaturization. It offers the opportunities for better diagonsis, pevention and treatment of human diseases. Besides, It aslo can be discovered the application in the field of food safety and counter-terrorism. Electrochemiacal DNA biosensor is a device composed of an active biomolecules with a signal transducer (electrodes). However, the mechanim of interaction between biomolecules and the solid electrodes is not yet clarified very well, which is the key question for the degin of the DNA biosensors with high performance. In this project, the ZnO/diamond electrodes with different shapes, sizes, dimensions, surface conditions, microstructures and crystal quality are deposited by microwave plasmar enhanced CVD techniques, sol-gel tehnique and hydrothermal synthesis. This project will focus on the influence of the nature of eletrodes on the functionalization of the electrode surface, DNA immobilizaiton techniques and electrochemical properties.
