中文摘要: 鸭坦布苏病毒(Duck Tembusu virus, DTMUV)病于2010年4月首先在我国上海、浙江等地爆发。该病的血清学、病原学等主要诊断方法已经成熟,而DTMUV与宿主先天性免疫系统的相互作用还不清楚。前期研究发现鸭RIG-I和MDA5参与DTMUV诱导的宿主免疫反应。本项目拟采用siRNA或超表达RIG-I和MDA5的方法,利用已经建立的萤火虫荧光素酶报告基因系统、荧光定量RT-PCR等技术,阐明RIG-I和MDA5如何参与DTMUV诱导的免疫反应,并确定其是否具有抗病毒感染功能。进一步在细胞中分别过表达DTMUV七个非结构蛋白和RIG-I或MDA5的效应结构域,利用上述技术揭示哪个/些非结构蛋白通过何种方式阻断RIG-I和MDA5介导的信号通路,从而逃避宿主的免疫监视。这将为深入理解DTMUV感染与机体免疫机理及筛选新型抗DTMUV疫苗的免疫佐剂提供科学依据。

英文摘要: The infectious disease caused by duck Tembusu virus (DTMUV) was first broken out in Shanghai, Zhejiang in April 2010. Although the diagnostic methods of serology and etiology have been established, the interaction between DTMUV and host innate immune system was still unclear. Our previous studies have demonstrated that RIG-I and MDA5 were involved in the immune response to DTMUV in ducks. In the present proposal, the method of over-expression or siRNA of RIG-I and MDA5, and the technique of firefly luciferase reporter system and real-time quantitative RT-PCR will be used to determine the roles of two receptors in DTMUV induced host innate immune and the ability of them in anti-DTMUV infection. Further study will investigate the roles of non-structural proteins of DTMUV in inhibiting RIG-I and MDA5-mediated signaling pathways to evade the host's immune surveillance by over-expressed of the seven non-structural proteins of DTMUV and effector domain of RIG-I and MDA5 in vitro. This study will be useful to further elucidate the pathogenesis of DTMUV and the mechanism of host immunity and provide a scientific basis for screening of vaccine adjuvants.
