中文摘要: DNA损伤造成基因组的不稳定和突变,是恶性肿瘤的基本特征之一。特异性靶向DNA损伤修复通路中的一些关键信号分子是抗肿瘤药物研发的一个重要方向。天然产物结构新颖生物学活性多样,在其众多的生物活性中,抗肿瘤活性是很重要的一个研究内容。近年的研究发现藤黄属植物中分离纯化的多种天然化合物有较强的抗肿瘤活性,但是具体的分子作用机制和靶点还很不清楚。我们在前期研究中以DNA损伤应答早期蛋白磷酸化H2AX为标记筛选了部分具有抗肿瘤活性的藤黄属天然化合物,发现它们能在抑制DNA损伤修复的同时诱导DNA双链断裂损伤和细胞自噬。本研究项目将在上述基础上,以DNA损伤应答,损伤修复和细胞自噬为研究方向,系统深入的探讨化合物的作用靶点和作用机制,为天然化合物抗肿瘤药物的研究提供重要的理论依据和潜在的临床价值。

英文摘要: DNA damage induces genome instability, which is the major hallmark of cancer. Targeting DNA repair proteins has been explored as a useful strategy in anti-cancer drug discovery. Natural products are rapidly gaining attention as sources of new drugs, particularly anti-cancer drugs. Recent studies have demonstrated that natural compounds purified from Garcinia species have potential anti-cancer activities, but the molecular mechanisms are poorly understood. Using DNA damage response protein phospho-H2AX as a marker, we screened natural compounds from Garcinia species. We found that some compounds can significantly inhibit DNA repair, induce DNA double strand breaks and autophagy. In this study, we will focus on DNA damage response, DNA repair and autophagy signaling pathways to identify the molecular mechanism and the potential targets of the natural compounds. It is anticipated that more natural compounds will be identified and clinically used as effective anti-cancer drugs for various cancer.
