中文摘要: 本课题将以体外模型为基础,从难于完全分解的耐受性核酸的角度研究食用核酸类物质的消化、吸收情况。课题将以单一纯核酸化合物及核酸含量较高的食材作为原材料,采用适当消化酶和生化条件,建立适用于食用核酸类成分的体外消化模型,利用其分析核酸消化降解规律和产物特点,探寻确定难于完全降解的消化耐受性核酸。主要消化耐受性核酸以Caco-2细胞模型研究其肠吸收情况,以人体血液和淋巴液为基质,研究其体液中的稳定性,以评价何种结构或状态的寡核苷酸可以在体内环境下"幸存"并吸收,为耐受性核酸在体内留存并进入组织和细胞的可能性提供依据。 课题创新之处在于:(1)将创造性的把体外模型引入食用核酸的研究中,为核酸研究另辟蹊径;(2)将首次针对可能尚有遗传表达生理功能的耐受性核酸,对其产生机制、结构特点、吸收特性及稳定性进行较系统研究。成果将对核酸摄入营养、安全,转基因食品、基因药物等核酸利用,提供技术支持和理论依据。

英文摘要: The project will study on the digestion and uptake of tolerant nucleic acids which are difficult to hydrolyze totally based on the in vitro model. Pure nucleic acids and food with higher nucleic acids are selected as research materials. Digestion enzyme and other biochemistry conditions will be tested to set up an in vitro digestion model which fits for edible nucleic acids. The digestion rule of nucleic acids and the characteristics of degradation products will be studied using this in vitro digestion model. Then the tolerant nucleic acids which show some resistance to digestion will be figured out. After that, digestion tolerant nucleic acids will be applied to Caco-2 cell model to investigate their intestine uptake. Their in vivo stability will also be studied using blood and lymph. The results will help us to find out that oligonucleotides with what kind of structures or sequences can survive from in vivo conditions and be uptaken by intestine. The conclusions of the research will also provide a basis for the possibilities of tolerant nucleic acid enter into human tissues or cells.
