中文摘要: 胰岛素抵抗(IR)是2型糖尿病核心特征之一。申请人前期研究发现,Hap1-Ahi1复合物参与了下丘脑胰岛素介导的摄食行为和能量平衡,该复合物可作为5-HT2C受体的抑制剂介导糖尿病、IR和肥胖的发生,提出Hap1-Ahi1信号通路是2型糖尿病发生发展的重要新机制(The Journal of Biological Chemistry. 2012)。桂皮醛(CA)为治疗糖尿病常用中药肉桂的主要有效成分,最新研究证实,CA能够改善大脑中胰岛素敏感性,但其作用的分子机制还有待进一步研究。本项目拟在前期工作基础上,从整体、组织、细胞和分子水平,观察CA改善大脑胰岛素敏感性的作用,并筛选其作用靶分子,验证CA抑制Hap1-Ahi1的体内信号转导机制。通过以上研究,不仅有助于阐明CA调节下丘脑Hap1-Ahi1通路的分子机制,也为中药抗糖尿病研究提供新思路,为研发治疗糖尿病的创新中药奠定基础。

英文摘要: Type 2 diabetes mellitus (T2DM) is mostly characterized by insulin resistance (IR). The applicant’s previous researches revealed that Hap1-Ahi1 compound in hypothalamus mediates feeding behavior and energy metabolism through insulin, thus may induce diabetes mellitus, IR and obesity. This indicates that Hap1-Ahi1 signaling pathway is an important new mechanism in the development of T2DM. (The Journal of Biological Chemistry. 2012) Cinnamaldehyde (CA),a major component of cinnamon extract,has been proven to regulate insulin sensitivity in human brains, of which the molecular mechanism requires further study. Based on previous researches, this project is designed to observe the regulating effects of CA on insulin sensitivity, screen the target sites, thus to validate the effects of CA on Hap1-Ahi1 signaling pathway. This study would help to reveal the molecular mechanism of CA on hypothalamic Hap1-Ahi1 signaling pathway regulation, providing new thinking of research ideas of chinese medicine in treatment of DM, and foundation of developing innovative herbal drugs.
