中文摘要: 选育提高猪肌内脂肪含量的关键是找到肌内脂肪沉积的组织特异性调控机制。P160家族基因通过调节PPARs等基因的活性从而对脂肪细胞分化和脂质代谢水平的高低发挥着精细调控的作用,因此P160家族基因表达和调控的组织差异性对机体脂肪差异分布的形成具有重要的调控作用。我们前期的研究工作发现P160家族基因的表达与肌内脂肪含量相关,并首次克隆了猪P160家族三个基因的全序列。本研究拟从基因的组织表达水平、对关键生理调控信号的反应性、基因转录调控因子水平、DNA甲基化水平以及miRNA 水平等角度,探讨P160家族NCOA1、NCOA2、NCOA3基因的表达和调控在肌内脂肪组织与皮下脂肪组织之间的差异,进而分析P160家族基因表达和调控的组织差异性对个体之间肌内脂肪与皮下脂肪差异性分布的作用机理,从而阐明P160家族基因组织特异性调控猪肌内脂肪沉积的分子机制。

英文摘要: The key to improving intramuscular fat (IMF)content is to discover the tissue-specific mechanism that regulates intramuscular fat deposition. By finely controlling the activity of the PPARs genes, P160 family genes regulate the the levels of adipocyte differentiation and lipid metabolism. Hence, the tissue-specific expression and regulation of P160 family genes play important roles in regulating body fat distribution in different tissues. In our previous research, we cloned pig P160 family gene sequences, and found that P160 family genes' expression correlated well with intramuscular fat content. In this study, we intend to investigate the differences between intramuscular fat and subcutaneous fat in P160 family genes' tissue expression, transcriptional factors levels, DNA methylation and miRNA levels, as well as the genes' reaction sensitivity to physiological regulatory signals. The effects of P160 gene family on the fat distribution will be studied by correlating the tissue differences in P160 expression and regulation with the ratio of intramuscular fat content to backfat thickness (IMF/BFT). This study will contribute to revealing the molecular mechanisms of P160 family genes in tissue-specific regulating the deposition of intramuscular fat in pigs.
