中文摘要: 花生四烯酸对海水鱼的繁育性能具有极为重要的调节作用,但其调节机制研究却几乎空白。本项目拟以我国重要的海水养殖种类大菱鲆为研究对象,采用梯度营养素投喂法对花生四烯酸调节海水鱼性类固醇激素合成的机制进行研究:首先,通过酶联免疫吸附技术研究花生四烯酸的中间代谢产物在性腺中的表达,并通过相关性分析初步比较这些中间产物在类固醇激素合成中的不同作用;其次,选取关键蛋白为指标运用分子生物学手段进一步探讨花生四烯酸及其代谢产物对类固醇激素合成三个关键环节(即对促性腺激素的响应、胆固醇原料的获得及合成过程酶促反应)的影响;同时,通过对性腺发育前期(II期)、中后期(III-IV期)、末期(V-VI期)的比较研究来揭示花生四烯酸对类固醇激素合成的调控作用在性腺发育不同时期间的动态变化规律。研究结果对揭示花生四烯酸调控海水鱼类固醇激素合成的机制具有重要推进作用,有助于更好地通过营养学手段人工调控海水鱼繁育性能。

英文摘要: Arachidonic acid plays a key role in reproduction of marine fish. However, little information has been known related to the mechanism involved in the regulation of arachidonic acid on marine fish reproduction. The present project was aimed to investigate the regulation of sex steroid hormones synthesis by dietary arachidonic acid with a feeding experiment on turbot broodstock. Firstly, this project examines the expression of different metabolites of arachidonic acid in gonad of experimental fish with enzyme-linked immunosorbent assay and compares the effects of these metabolites on sex steroid hormone synthesis via correlation analysis. Secondly, the effects of dietary arachidonic acid on characteristic proteins in three key steps of sex steroid hormone synthesis, i.e., response to gonadotrophin, obtaining of the cholestoral substrate and enzymatic reaction of synthesis process, were studied with molecular biological method. Meanwhile, the regulation of sex steroid hormone synthesis by dietary arachidonic acid in different developmental stages of gonad, i.e., early stage (stage II), mid stage (stage III-IV) and later stage (stage V-VI), were also studied in order to elucidate the dynamic variation of this regulation with developmental stage.
